Effects of temperature and metallic cations on the combination of interferon inducer polyinosinic polycytidylic acid (polyI:C)

    Abstract:

    Effects of temperature and metallic cations on the synthesis of polyinosinic polycytidylic acid were tested to improve its structure. The results showed significant hypochromic effects of polyI:C formed at 45℃,which indicated that the best combination of polyI:C were achieved at this temperature, while the metallic cations including Na+、Ca2+、Mg2+、Mn2+ and Zn2+ could enhance the polyI:C combination. 0085mol/L PolyI:C combination achieved saturation when Na+ concentration was, and the divalent cations including Ca2+、Mg2+、Mn2+ and Zn2+ played positive effect on the combination. However, Cu2+ tend to destroy the double stranded structure. It is suggested that polyI:C should be made at 45℃ and suitable concentration of Na+ and Ca2+should be added to make its structure more tight in order to avoid RNase’s digesting. This result is useful for synthesizing polyI:C at a high quality.
